[Objective]To understand the relationship between Oleosin gene family and drought tolerance in foxtail millet,and explore gene related to drought tolerance.[Methods]In this study,drought-tolerant variety Gou Gou Mu Ji Ju(GG)and drought-sensitive variety Jinfen 16(JF16)were used as experimental materials.They were treated by PEG at seedling for simulating drought stress and expression analysis was performed.In order to understand the Oleosin gene family in foxtail millet,bioinformatics analysis was conducted.[Results]The result showed that the Oleosin gene(Seita.9G579700)was sig-nificantly up-regulated in both GG and JF16 under drought stress.The Oleosin gene family was small and had only 6 mem-bers with molecular weight of 15~18 kDa.Phylogenetic tree analysis showed that Oleosin family in foxtail millet were clus-tered with monocotyledonous,such as maize,sorghum and rice,but the relationship with dicotyledonous A rabidopsis thali-ana was significant distant.The analysis of promoter elements showed that the six Oleosin genes in foxtail millet contained light response,ABA response,MeJA response,and MYB binding sites which were associated with drought tolerance.[Con-clusion]It could be proposed that the Oleosin gene was a candidate gene for regulating drought stress response in foxtail mil-let.Oleosin gene may be induced by drought or ABA,and MeJA.This study could provide a theoretical reference for further research on the understanding the function of Oleosin genes and its relationship with drought tolerance in foxtail millet.
