Extraction and content comparison of 1-deoxynojirimycin from mulberry leaves
This study mainly explores the extraction conditions of 1-deoxynojirimycin(DNJ)from mulberry leaves.Determination of 1-deoxynojirimycin(DNJ)content using high-performance liquid chromatography ultraviolet detection method from superior mulberry tree varieties inYuncheng City.Mulberry leaves were ultrasonically extracted with 0.01 mol/L hydrochloric acid,and 1-deoxynojirimycin(DNJ)was derivatized with fluorene methoxycarbonyl chloride(FMOC-C1)in sodium borate buffer pH 8.5.-Visible detector detection.In this experiment,the ultrasonic extraction conditions were explored,and the extraction conditions were optimized by single factor test and orthogonal test,and the optimal extraction conditions were selected,that is,70 ℃,350 W ultrasonic extraction for 60 min.the peaks were well separated under these extraction conditions,with no tailing and no continuity.1-Deoxynojirimycin had a good linear relationship in the concentration range of 0.1-1.5 mg/mL(R2=0.9989).The content of 1-deoxynojirimycin in different varieties of mulberry leaves was different.The content of 1-deoxynojirimycin in red big leaves(8-12)was the highest(1.5456 mg/g),and the content of 1-deoxynojirimycin in Huanggelu(8-15)was the highest.1-Deoxynojirimycin had the lowest content(0.1125 mg/g).CONCLUSION:The method is simple to operate and suitable for extraction and content determination of 1-deoxynojirimycin in mulberry leaves.
