益肾泄浊方对肾纤维化大鼠的治疗作用及机制

目的 探讨益肾泄浊方对腺嘌呤所致的肾纤维化大鼠的作用及其机制.方法66只大鼠随机分为空白组、模型组、尿毒清组(1.6g/kg)、益肾泻浊方低剂量组(0.65 g/kg)、益肾泻浊方中剂量组(1.3 g/kg)、益肾泻浊方高剂量组(2.6 g/kg),每组各11只.除空白组外,其余各组采用腺嘌呤(200 mg/kg)灌胃28 d诱导肾纤维化大鼠模型,造模后,尿毒清组和益肾泄浊方组给药治疗8周.测定SCr和BUN含量、24 h UTP和24 h mAlb水平;HE和Masson染色镜下观察肾脏组织病理变化;免疫组化测大鼠肾脏组织分泌型糖蛋白(Wnt1)、β-连环蛋白(β-catenin)的蛋白表达情况.结果 与空白组比较,模型组、尿毒清组、低剂量组、中剂量组和高剂量组24 h UTP和24 h mAlb、SCr和BUN含量、Wnt1和β-catenin蛋白表达均升高(P<0.05);与模型组比较,尿毒清组、低剂量组、中剂量组和高剂量组24 h UTP和24 h mAlb、SCr和BUN水平、Wnt1和β-catenin蛋白表达均降低(P<0.05);与尿毒清组比较,低剂量组24 h UTP和24 h mAlb、SCr和BUN水平差异无统计学意义(P>0.05),Wnt1和β-catenin蛋白表达均降低(P<0.05),中剂量组和高剂量组24 h UTP和24 h mAlb、SCr和BUN水平、Wnt1和β-catenin蛋白表达均降低(P<0.05);与低剂量组比较,中剂量组和高剂量组24 h UTP和24 h mAlb、SCr和BUN水平、Wnt1和β-catenin蛋白表达均降低(P<0.05);与中剂量组比较,高剂量组24 h UTP和24 h mAlb、SCr和BUN水平、Wnt1和β-catenin蛋白表达均降低(P<0.05).结论 益肾泄浊方能够通过调控Wnt/β-catenin信号通路来阻抑肾纤维化的发展,从而起到保护肾脏的作用.
Therapeutic effect and its mechanism of Yishen Xiezhuo decoction on renal fibrosis in rats
Objective To observe the regulatory effect of Yishen Xiezhuo decoction on adenine-induced renal fibrosis in rats and explore its underlying mechanisms.Methods Sixty-six rats were randomly divided into blank group,model group,Niaoduqing group(1.6 g/kg),low dose group(0.65 g/kg Yishen Xiezhuo decoction),medium dose group(1.3 g/kg Yishen Xiezhuo decoction),and high dose group(2.6 g/kg Yishen Xiezhuo decoction),with 11 rats in each group.Except for blank group,the rats in the other groups were orally administrated with adenine(200 mg/kg)to induce the renal fibrosis for 28 d.After modeling,the rats in Niaoduqing group and Yishen Xiezhuo decoction groups were treated with medication for 8 weeks.The levels of SCr and BUN,24 h UTP and 24 h mAlb were determined.Pathological changes of renal tissues were observed by HE and Masson staining.Immunohistochemistry was performed to detect the protein expression of secreted frizzled-related protein(Wnt1)and β-catenin in rat renal tissues.Results Compared with blank group,the levels of 24 h UTP,24 h mAlb,SCr and BUN,and the expressions of Wnt1 and β-catenin proteins were increased in model group,Niaoduqing group,low dose group,medium dose group and high dose group(P<0.05).Compared with model group,the levels of 24 h UTP,24 h mAlb,SCr and BUN,and the expressions of Wnt1 and β-catenin proteins were decreased in Niaoduqing group,low dose group,medium dose group and high dose group(P<0.05).Compared with Niaoduqing group,24 h UTP and 24 h mAlb,SCr and BUN levels showed no significant changes in low dose group(P>0.05),but the expressions of Wnt1 and β-catenin proteins were decreased(P<0.05).Compared with Niaoduqing group,the levels of 24 h UTP,24 h mAlb,SCr and BUN,and the expressions of Wnt1 and β-catenin proteins were decreased in medium dose group and high dose group(P<0.05).Compared with low dose group,the levels of 24 h UTP,24 h mAlb,SCr and BUN,and the expressions of Wnt1 and β-catenin proteins were decreased in medium dose group and high dose group(P<0.05).Compared with medium dose group,the levels of 24 h UTP,24 h mAlb,SCr and BUN,and the expressions of Wnt1 and β-catenin proteins were decreased in high dose group(P<0.05).Conclusion Yishen Xiezhuo decoction can inhibit the development of renal fibrosis by modulating the Wnt/β-catenin signaling pathway,thereby exerting a protective effect on the kidneys.
