Echocardiography assessment of Kawasaki disease mouse model
Objective To observe the changes of coronary artery imaging and cardiac function in the murine model of LCWE-induced Kawasaki disease(KD)using high frequency ultrasound system and provide a basis for the preparation of murine KD model and its function assessment.Methods Fifty mice were randomly divided into two groups:model group(LCWE-induced KD model,n=30)and control group(no treatment,n=20).The coronary artery dimensions were measured at day 15,21 and 30 after modeling by small animal ultrasound,left ventricular end-diastolic diameter(LVEDD),left ventricular end-systolic diameter(LVESD),left ventricular ejection fraction(LVEF)and left ventricular short axis shortening rate(LVFS)were recorded by M-mode echocardiography.One mouse in control group and model group was killed at day 15,21,30 to collect the heart samples for pathological examination,respectively.Results Echocardiography showed a high density echo around the coronary artery wall in model group at day 15,21 after modeling,and diffuse thickening at day 30 after modeling.The diameters of both left and right coronary arteries in model group were larger than those in control group at day 21(P<0.05);however,there was no significant difference in the inner diameter of coronary artery between two groups at day 15 and 30 after modeling(P>0.05).LVEDD,LVESD,LVEF and LVFS were lower in model group than in control group at day 15 after modeling(P<0.05),and LVEF and LVFS were lower at day 21(P<0.05),and LVEDD,LVEF and LVFS were lower at day 30(P<0.05).At day 15 after modeling,the interstitial edema of heart,mild lymphocyte infiltration and diffuse mild hyperplasia of fibrous connective tissue on the epicardium were found,and the myocardial hyperplasia accompanied by calcium deposition was also detected.At day 21 after modeling,the epicardium of KD mouse was found to be infiltrated by a large number of lymphocytes,eosinophils and monocytes.At day 30 after modeling,the coronary artery of KD mouse presented thickened wall,discontinuous elastic fibers,slightly thickened intima,and local hyaline degeneration.Conclusion The injury of coronary artery in the murine model of KD induced by LCWE is similar with the natural pathology progression of KD children,and it can be used to simulate the changes of coronary artery and cardiac function of KD.
