Optimization of extraction process of Shuxin Anshen ointment based on response surface method and UPLC-MS/MS
Objective To optimize the extraction process of Shuxin Anshen ointment by response surface method.Methods With the contents of monoxide,maggianin,paeoniflorin,spinoside,6-gingerol simultaneously determined by ultra-high performance liquid chromatography,the extraction rate,and the total polysaccharide content as the comprehensive score indexes,the effects of water adding ratio,extraction time,and extraction times on the extraction process of Shuxin Anshen ointment were investigated by a single-factor test.Based on the single factor results,the extraction process of Shuxin Anshen ointment was optimized by response surface method.Results After single factor test and response surface method optimization,the optimal process conditions were as follows:adding 10 times water,extracting twice,and extracting 90 min each time.Under this condition,the RSD of the predicted comprehen-sive score and the measured score was 0.82%.Conclusion The extraction process of Shuxin Anshen ointment optimized by the response surface method is reasonable and feasible,which can provide an experimental reference for its subsequent development and research.
