Expression and prognostic value of UBE2L6 in ovarian cancer
Objective To explore the expression of UBE2L6 gene in ovarian cancer and its relationship with prognosis by bioinformatics method.Methods A total of 150 patients with ovarian cancer hospitalized in Shanxi Bethune Hospital from July 2014 to February 2022 and 25 patients undergoing total hysterectomy plus bilateral adenectomy due to adenomyosis were collected.Immunohistochemical SP method was used to detect the expression of UBE2L6 protein.The expression of UBE2L6 protein was compared between ovarian cancer and normal ovarian tissue.The relationships between UBE2L6 protein expression and clinicopathological features of ovarian cancer patients were analyzed.Independent risk factors affecting progression-free survival of ovarian cancer patients were screened byX2 test and COX risk regression analysis,and survival curves were plotted by Kaplan-Meier method.The correlation between UBE2L6 expression and clinicopathological indicators of ovarian cancer was analyzed based on online gene expression database,and the correla-tion between UBE2L6 expression and prognosis of ovarian cancer was retrieved through Kaplan-Meier database.Kyoto Encyclopedia of Genes and Genomes enrichment analysis was performed to predict gene function.The correlation between high expression of UBE2L6 in ovarian cancer and local immune invasion was analyzed through the Tumor Immune Estimation Resource(TIMER)database.Results Compared with normal ovarian tissues,UBE2L6 expression was increased in ovarian cancer tissues(P<0.05),and it was associated with the poor prognosis.COX regression analysis showed that UBE2L6 high expression was an independent risk factor for progression-free survival of ovarian cancer patients(P<0.05).Gene enrichment analysis showed that UBE2L6 may affect the development and prognosis of ovarian cancer through inflammatory pathways.The TIMER database showed that the expression level of UBE2L6 was posi-tively correlated with B cells(P=9.87 ×10-11),CD8+T cells(P=9.64 ×10-13),neutrophils(P=2.13×10-22),dendritic cells(P=6.50 × 10-15),CD4+T cells(P=3.51 x 10-4),and negatively correlated with tumor purity(P=7 × 10-8).Conclusion UBE2L6 may lead to poor prognosis by affecting immune cells,which may be a potential target for ovarian cancer treatment.
