Effect of TRIM59 on proliferation,apoptosis,invasion and migration of human hypopharyngeal cancer cells
Objective To investigate the effects of tripartite motif containing 59(TRIM59)on the biological behaviors of hypopharyngeal cancer cells Fadu and its mechanism.Methods The expression of TRIM59 in different types of cancer was investigated through the Tumor Immune Estimation Resource database 2.0(TIMER2),and its expression difference in head and neck tumors was initially eva-luated.Nasopharyngeal carcinoma Fadu cells were cultured and randomly divided into experimental group(si-TRIM59-1 group,si-TRIM59-2 group,si-TRIM59-3 group)and control group(NC group).Real-time quantitative PCR was used to detect the transfection efficiency,and the two groups with successful transfection and the highest efficiency of gene downregulation were selected for subse-quent experiments.The proliferation ability of Fadu cells was detected by CCK-8 assay and colony formation assay.The migration and invasion abilities of Fadu cells were detected by Transwell assay and cell scratch assay.After TRIM59 was downregulated,the expressions of related proteins(TRIM59,JAK2/p-JAK2,STAT3/p-STAT3,Bcl-2,Bax)were detected by Western blot assay.The apoptosis rate of Fadu cells was detected by flow cytometry.Results Compared with NC group,the relative mRNA expression of TRIM59 was decreased in si-TRIM59-1 group,si-TRIM59-2 group and si-TRIM59-3 group(P<0.001),especially si-TRIM59-1 group and si-TRIM59-3 group(P<0.05).So si-TRIM59-1 and si-TRIM59-3 were chosen for subsequent experiments.Compared with NC group,the proliferation,migration and invasion abilities of Fadu cells in si-TRIM59-1 group and si-TRIM59-3 group were decreased(P<0.05).Compared with NC group,the expression levels of TRIM59,JAK2/p-JAK2,STAT3/p-STAT3 were decreased in si-TRIM59-1 group and si-TRIM59-3 group(P<0.05),and the expression of inhibitory protein Bcl-2 was decreased while the expression of inducible protein Bax was increased(P<0.05).Compared with NC group,the apoptosis rate was increased in si-TRIM59-1 group and si-TRIM59-3 group(P<0.05).Conclusion TRIM59 downregulation can inhibit the proliferation,migration and invasion of human hypopharyngeal cancer cell line Fadu,and induce the apoptosis rate,which may be related to the JAK2/STAT3 pathway.
