Effects of astragaloside on sex hormone levels and ovarian senescence in mice with polycystic ovary syndrome
Objective To investigate the effects of astragaloside on the levels of sex hormone and senescence of ovary tissues in mice with polycystic ovary syndrome(PCOS).Methods Thirty female C57BL/6 mice were randomly divided into normal control group,model group and astragaloside group,with 10 mice in each group.The PCOS model was induced by subcutaneous injection of dehydro-epiandrosterone(DHEA,70 mg/kg)on the nape back combined with intraperitoneal injection of D-galactose(200 mg/kg)once daily for 21 consecutive days.The mice in AST group were orally administered with AST(50 mg/kg)once a day for 4 consecutive weeks after the PCOS mice were sucessfully established.During the last 10 days of administration,the vaginal smear was performed to assess the estrus cycle.After the administration,the ovarian index was calculated.HE staining was conducted to observe the histopathological changes of the ovary.Serum levels of sex hormones testosterone(T),estradiol(E2),luteinizing hormone(LH)and follicle-stimulating hormone(FSH)were measured by ELISA.The protein levels of GLB1,P16,and p-P65 in ovarian tissues were detected by immunohis-tochemistry.The protein levels of GLB1,P16,p-P65 and P65 in ovarian tissues were detected by Western blot.Results Compared with control group,the oestrous cycle of mice was disorder in model group,the ovarian index was elevated(P<0.05),and the ovary was polycystic;the serum levels of T and LH were significantly enhanced(P<0.05),while the contents of E2 and FSH were signifi-cantly declined(P<0.05);and the levels of GLB1,P16 and p-P65 were augmented(P<0.05).Compared with PCOS group,the oestrous cycle of mice was restored in AST group,and the ovarian index was decreased(P<0.05);the serum levels of T and LH were significantly decreased(P<0.05),while the contents of E2 and FSH were significantly increased(P<0.05);and the levels of GLB1,P16,and p-P65 were reduced(P<0.05).The level of P65 showed no significant difference between groups.Conclusion AST can effectively balance the levels of sex hormone in PCOS mice and relieve the senescence of ovary tissues,which may be related to the inhibition of NF-κB signaling pathway.
