Causal relationship between body composition and calcaneal bone mineral density:a Mendelian randomization analysis
Objective To investigate the causal relationship between body composition and calcaneal BMD by Mendelian randomiza-tion methods.Methods The genome-wide association study data were used.The exposure factors were whole body fat mass,whole body water mass,and whole body fat free mass.The outcome variable was calcaneus BMD.IVW method,MR-Egger method,weighted median method,simple mode method and weighted mode method were used for Mendelian randomization analysis.Heterogeneity,sen-sitivity and pleiotropy were tested to evaluate the reliability.Results The IVW method showed a positive causal relationship between whole body fat mass and calcaneus BMD(β=1.10,95%CI 1.05-1.15,P<0.001).The results of MR-Egger method,weighted median method and weighted mode method were consistent with that of IVW method.Sensitivity analysis results showed no heterogeneity or horizontal pleiotropy of single nucleotide polymorphisms(SNP).IVW method showed no significant causal relationship between whole body water mass and calcaneus BMD(β=0.96,95%CI 0.91-1.02,P>0.05).IVW method showed no significant causal relationship between whole body fat free mass and calcaneus BMD(β=0.97,95%CI 0.91-1.03,P>0.05).Conclusion There is a positive causal relationship between whole body fat mass and calcaneal bone mineral density.
