Effect of platelet-rich plasma microspheres on urodynamics and collagen metabolism of anterior vaginal wall in rats with urinary incontinence after radical cervical cancer surgery
Objective:To investigate the effects of platelet-rich plasma microspheres on urodynamics and colla-gen metabolism of anterior vaginal wall in rats with urinary incontinence after radical cervical cancer surgery.Methods:A total of 60 female rats that had not given birth for 6 to 8 weeks were divided into normal group,model group,platelet-rich plasma group,blank gelatin microsphere group and platelet-rich plasma microsphere group with 12 rats in each group according to random number table method.After successful preparation of urinary incontinence rat model after radical cervical cancer surgery,normal group and model group were fed normally,and the other three groups were injected with platelet-rich plasma,blank gelatin microspheres and platelet-rich plasma microspheres with microsyringes.The general condition,urinary dynamic indexes,internal bladder pressure,urethral sphincter tissue morphology,anterior vaginal wall transforming growth factor-β(TGF-β),tissue inhibitor of metalloproteinase-1(TIMP-1),matrix metalloproteinase-1(MMP-1),matrix metalloproteinase-9(MMP-9),type Ⅰ collagen(Col-ⅠmRNA expression level,TGF-β,TIMP-1,MMP-1,MMP-9,Col-Ⅰ protein expression.were observed in each group.Results:Compared with the model group,urinary motility indexes,bladder internal pressure,TGF-β mRNA and pro-tein,TIMP-1 mRNA and protein,MMP-1 mRNA and protein,MMP-9 mRNA and protein,Col-Ⅰ mRNA and pro-tein in normal group,platelet-rich plasma group,blank gelatin microsphere group and platelet-rich plasma micro-sphere group mRNA and protein were improved(all P<0.05).Compared with platelet-rich plasma group and blank gelatin microsphere group,urinary motility index,bladder internal pressure,TGF-β mRNA and protein,TIMP-1 mRNA and protein,MMP-1 mRNA and protein,MMP-9 mRNA and protein,Col-Ⅰ mRNA and protein were im-proved in normal group and platelet-rich plasma micropellet group(all P<0.05).Compared with platelet-rich plas-ma micropellet group,urinary motility index,bladder pressure,TGF-β mRNA and protein,TIMP-1 mRNA and pro-tein,MMP-1 mRNA and protein,MMP-9 mRNA and protein,Col-Ⅰ mRNA and protein in normal group were im-proved(all P<0.05).The above indexes in platelet-rich plasma group were not different from blank gelatin micro-sphere group(all P>0.05).Conclusion:Platelet-rich plasma microspheres can restore urodynamics and regulate col-lagen metabolism in rats with urinary incontinence after radical cervical cancer surgery.
