Effect of Trehalose on Seed Germination and Seedling Physiology of Polygala Tenuifolia Under Drought and Salt Stress
Objective To investigate the physiological effects of trehalose on seed germination and seedlings of Polygala tenuifolia under drought and salt stress.Methods Polygala tenuifolia Willd is used as the research object,and the seed soaking experiment involved soaking Polygala tenuifolia Willd seeds in a trehalose solution with a concentration of 5~25 mmol·L-1 for 24 hours,and then placing them on a germination bed treated with 224 mg·mL-1 sodium chloride(NaCl)and 3810 mg·mL-1 polyethylene glycol(PEG-6000);The seed mixing experiment was conducted by washing the seeds of Polygala tenuifolia with running water for 24 hours,and then placing them in a germination bed treated with NaCl and PEG-6000 supplemented with 5~25 mmol·L-1 trehalose solution.Observe continuously for 15 days and record relevant data such as seed germination rate,root length,shoot length,and hypocotyl length.Results The appropriate concentration of trehalose has a certain effect on alleviating the germination and seedling growth of Polygala tenuifolia seeds under salt and drought stress.Under salt stress,the germination rate of Polygala tenuifolia Willd seeds was not significantly affected compared to the control group,but the seedling length was significantly shortened.The 10 mmol·L-1 trehalose treatment group under soaking treatment and the 5 mmol·L-1 trehalose treatment group under seed mixing treatment had the most significant promoting effect on seedlings.Drought stress can inhibit the germination of Eucommia ulmoides seeds and the growth of seedlings.Both soaking and seed mixing treatments showed the highest germination rate in the 15 mmol·L-1 trehalose treatment group,while the 5 mmol·L-1 trehalose treatment group had the best promoting effect on seedling growth.Soaking and seed mixing with trehalose can activate the antioxidant system in the seedlings of Polygala tenuifolia to a certain extent,and enhance the activities of catalase(CAT)and peroxidase(POD)to cope with salt stress and drought stress environments.Conclusion Soaking or mixing seeds with appropriate concentrations of trehalose can alleviate the inhibitory effects of salt stress and drought stress on the germination of Eucommia ulmoides seeds,promote the growth of embryonic roots,and enhance the antioxidant enzyme activity of seedlings.This can provide a reference for Eucommia ulmoides planting under adverse stress.
