Correlation of BRCA1 Gene with Immune Cell Infiltration and Prognosis in Lung Adenocarcinoma
Objective To discuss the correlation of breast cancer susceptibility gene 1(BRCA1)gene with immune cell infiltration and prognosis in lung adenocarcinoma(LU AD).Methods The pairs of specimens of lung tumor tissues and adjacent normal lung tissues(>10cm)resected from 68 patients with LUAD were collected.The quantitative real time polymerase chain re-action(qRT-PCR)was used to detect the mRNA expression level of BRCA1,and immunohistochemical method was adopted to de-tect the gene expression level of BRCA1.Then the correlation of BRCA1 gene with immune cell infiltration and prognosis in LUAD was verified via Oncomine,Kaplan-meier Plotter and Tumor Immune Estimation Resource(TIMER)database.Results Oncomine database analysis showed that BRCA1 gene expression level in LUAD tissues was higher than that in adjacent tissues,with statisti-cal difference(P<0.05).The qRT-PCR analysis showed that the BRCA1 mRNA expression level in LUAD tissues was higher than that in adjacent tissues,with statistical difference(P<0.05).TIMER database analysis indicated that BRCA1 expression was closely correlated with the infiltration level of neutrophils(γ=0.147,P=1.28 ×10-3),but was not significantly correlated with the infiltration level of B cells(γ=-0.09,P=4.73 × 10-2),CD8+T cells(γ=0.047,P=3.00 × 10-1),CD4+T cells(γ=0.039,P=3.91 × 10-1),macrophages(γ=-0.038,P=4.05 × 10-1)and dendritic cells(γ=0.039,P=3.85 × 10-1).Kaplan-meier Plotter database analysis suggested that the overall survival and disease-free survival rates of LUAD patients with high expression of BRCA1 gene were lower than those with low expression of BRCA1 gene,with statistical difference(P<0.05).Conclusion BRCA1 gene and mRNA are highly expressed in LUAD tissues,and the BRCA1 gene is closely related to poor prog-nosis and neutrophil infiltration in LUAD patients.
