Mechanism of miR-384 Targeting HMGA2 on the Growth,Metastasis and Invasion of Hepatocellular Carcinoma Cells
Objective To investigate the effect of miR-384 targeting high mobility protein A2(HMGA2)on the prolifera-tion,migration and invasion of liver cancer cells and its mechanism.Methods HUH7 cells in logarithmic growth phase were di-vided into the control group,miR-384 NC group and miR-384 mimic group.qRT-PCR was used to detect the expression of miR-384,MTT method was used to detect cell proliferation,Transwell assay was used to detect cell migration and invasion,and dual lu-ciferase gene reporter method was used to detect the targeting relationship.The relative expressions of HMGA2,E-cad,N-cad and Vimentin were detected by Western blotting.Results The expression level of miR-384 in human liver cancer cells HEP3 B,BEL-7402 and HUH7 was lower than that in human liver normal epithelial cells THLE3.There were no significant differences in the expression level of miR-384,cell survival rate,the number of migrating and invading cells,and the relative expression levels of HMGA2,E-cad,N-cad and Vimentin between the control group and the miR-384 NC group(P>0.05).Compared with the miR-384 NC group,the expression level of miR-384 in the miR-384 mimic group was increased,the cell survival rate was decreased,the number of migrating and invading cells was decreased,the relative expression of HMGA2,N-cad and Vimentin protein was de-creased,and the relative expression of E-cad protein was increased(P<0.05).Conclusion Overexpression of miR-384 can in-hibit the proliferation of hepatocellular carcinoma HUH7 cells and reduce the migration and invasion ability of cancer cells,possi-bly by reducing the expression of HMGA2 and regulating the expression of epithelial-mesenchymal transition(EMT)related pro-teins.
