miR-339-5p Affects the Metastatic Activity of Glioma Cells by Targeting and Regulating p53 Expression
Objective To investigate the effect of miR-339-5p on glioma U87 cells and to explore the related mecha-nism.Methods U87 cells were divided into 4 groups:blank group,miR-339-5p mimic group,si-p53 group,and miR-339-5p mimic+si-p53 group.The cell proliferation rate was detected by MTT assay;the invasion ability of U87 cells was detected by Tr-answell assay;the apoptosis rate was detected by flow cytometry;the targeting relationship between miR-339-5p and p53 was de-tected by luciferase reporter gene assay;the expression of miR-339-5p and p53 was analyzed by real-time fluorescence quantitative polymerase chain reaction and protein immunoblotting levels.Results The miR-339-5p expression levels of cells in the miR-339-5p mimic and miR-339-5p mimic+si-p53 groups were higher than those in the blank and si-p53 groups.p53 mRNA and protein levels of cells in the miR-339-5p mimic group were higher than those in the blank,si-p53 and miR-339-5p mimic+si-p53 group(P<0.05).miR-339-5p mimic and p53-WT cotransfected cells had significantly increased luciferase activity(P<0.05).miR-339-5p mimic group had lower cell proliferation rate and number of cell invasion than the blank group,si-p53 group and miR-339-5p mimic+si-p53 group,while the apoptosis rate in the miR-339-5p mimic group was higher than that in the blank group,si-p53 group and miR-339-5p mimic+si-p53 group(P<0.05).Conclusion miR-339-5p inhibits the proliferation and invasion of glioma U87 cells and promotes apoptosis of glioma U87 cells,while this process is associated with targeted activation of p53.
