Correlation Analysis of MED8 Expression and Clinicopathological Features and Prognosis in Hepatocellular Carcinoma
Objective To investigate the expression,the diagnostic and the prognostic value of mediator complex subunit 8(MED8)in hepatocellular carcinoma(HCC)as well as its possible mechanism in regulating HCC.Methods HCC RNASeq data and clinical information were downloaded from the TCGA database.The expression of MED8 in HCC cancer tissue and the paired paracancerous samples were analyzed.Western blot was used to detect the expression of MED8 in HepG2 cells and QSG-7701 normal liver cells.DESeq2 package and clusterProfiler package were used to analyze the expression difference and gene ontology(GO)enrichment analysis of the data in MED8 high and low expression groups.Immunohistochemistry was used to detect the ex-pression of the classical complement pathway-related protein C1qc and its relationship with MED8.The correlation and prognostic value of MED8 level with clinicopathological characteristics were analyzed by survival curve and ROC curve.Results MED8 was highly expressed in HCC cancer tissues and liver cancer cells.Enrichment analysis showed that proteins interacting with MED8 were mainly involved in biological processes such as complement activation and ion response;while the classical complement path-way-related protein C1qc was up-regulated in liver cancer samples and positively correlated with MED8 expression.High expres-sion of MED8 was closely related to T stage,pathological stage and histological grade of HCC.In addition,high levels of MED8 in HCC are positive correlation with poor overall survival and disease-related survival.Receiver Operating Characteristic(ROC)curve results indicate that the area under the curve(AUC)of MED8 is 0.817,indicating that MED8 may be a potential diagnostic molec-ular marker for HCC.Conclusion MED8 is closely related to the clinical stage and prognosis of HCC patients,and it may affect the prognosis of patients through the complement activation pathway.
