生物钟基因Bmal1、Per2在血液肿瘤细胞株Raji、Hut-78、OCI-LY8及HL-60中的昼夜表达规律

目的 通过检测生物钟基因Bmal1、Per2 在血液肿瘤细胞株Raji、Hut-78、OCI-LY8 及HL-60 细胞中不同时间点的mRNA表达情况,探讨其与血液肿瘤发生发展的可能关系.方法 血液肿瘤细胞株Raji、Hut-78、LY-8、HL-60 经血清休克法诱导节律后,RT-PCR法检测生物钟基因Bmal1 及Per2 在不同时间点的mRNA转录水平,以时间为变量分别对其转录量进行余弦函数拟合,通过余弦函数预测基因转录高峰及低谷时段.结果 Bmal1 和Per2 基因表达在OCI-LY8、Hut-78、HL-60 细胞中呈现昼夜节律(P<0.05);Raji细胞内Bmal1 基因无节律性表达(P>0.05),而Per2 基因呈昼夜节律表达(P<0.05).结论 生物钟基因Per2 和(或)Bmal1 在多种血液肿瘤细胞株内呈昼夜节律性表达,其可能参与血液肿瘤的发生发展.
The Expressional Circadian Rhythms of Clock Genes Bmal1 and Per2 in Raji、Hut-78、OCI-LY8 and HL-60 Cell Lines
Objective To detect the circadian rhythms of the expressional levels of Bmal1 and Per2 genes in hematologi-cal tumor cell lines Raji、Hut-78、OCI-LY8 and HL-60,and to explore the possible connections between them and the occurrence and development of hematological tumor.Methods The transcriptional levels of Bmal1 and Per2 at different times of the day in Raji、Hut-78、OCI-LY8 and HL-60 cell lines were measured by real-time PCR after the induction of synchronous rhythms of the clock gene expression.Cosinor analysis was performed by fitting transcription levels of genes at individual time points to predict the peak and trough periods of gene transcription in different cell lines.Results Circadian rhythms in the transcriptional level of Bmal1 and Per2 were observed in OCI-LY8、Hut-78、and HL-60 cell lines(P<0.05);Nocircadian rhythm was found in the tran-scriptional level of Bmal1 in Raji cell lines while the circadian rhythm of Per2 in it was found(P<0.05).Conclusion Circadi-an rhythms of the transcriptional levels of Bmal1 and/or Per2 could be found in various kinds of hematological tumor cell lines,in-dicating that it may take part in the occurrence and development of hematological tumors.
