Expression and Clinical Significance of CREB1 and CREB3 in Gastric Cancer and Pre-cancerous Lesions
Objective To investigate the expression and clinical significance of cAMP responsive element binding pro-tein 1(CREB1)and cAMP responsive element binding protein 3(CREB3)in gastric cancer and precancerous lesions.Methods 40 cases of chronic superficial gastritis,40 cases of chronic atrophic gastritis with intestinal metaplasia,40 cases of dysplasia,and 50 cases of gastric cancer were collected.The expressions of CREB1 and CREB3 in the 4 tissues were detected by immunohisto-chemical method,and the relationship between CREB3 and clinicopathological parameters was analyzed.A total of 30 cases of chronic superficial gastritis,chronic atrophic gastritis with intestinal metaplasia and gastric cancer were collected from fresh tissues during the same period.Western blot was used to detect the expression of CREB1 and CREB3 in different gastric tissues.Kaplan-Meier plotter was used to analyze the correlation between the expression of CREB1 and CREB3 and the overall survival time(OS)and progression-free survival time(FP)of gastric cancer patients.The position of CREB1 and CREB3 in signaling pathway and the upstream and downstream genes related to them were analyzed using STRING database.Results Immunohistochemistry showed that the positive expressions of CREB1 and CREB3 in gastric cancer group,dysplasia group,atrophic gastritis with intesti-nal metaplasia group were significantly higher than those in chronic superficial gastritis group(P<0.05).The positive expression rates of CREB1 and CREB3 in gastric cancer group were significantly higher than those in chronic atrophic gastritis with intestinal metaplasia(P<0.05),but there was no significant difference between them and dysplasia group(P>0.05).Analysis of clinico-pathological parameters showed that the expression levels of both were correlated with depth of invasion,TNM stage,vascular inva-sion and lymph node metastasis(P<0.05).There was no significant correlation with gender,age,tumor size and differentiation degree(P>0.05).Western blot results showed that the expression of CREB1 and CREB3 protein increased in chronic superficial gastritis,chronic atrophic gastritis with intestinal metaplasia and gastric cancer,with statistical significance(P<0.01).Kaplan-Meier plotter analysis showed that both OS and FP were shortened in gastric cancer patients with high CREB1 and CREB3 expres-sion.Conclusion CREB1 and CREB3 may be related to the occurrence,development and metastasis of gastric cancer,and the high expression of CREB1 and CREB3 may be related to the poor prognosis of gastric cancer patients.
