The Expression and Significance of LncRNA HULC in Papillary Thyroid Carcinoma Tissues
Objective To investigate the expression and significance of long chain non coding RNA(lncRNA)and high expression transcript of liver cancer(HULC)in papillary thyroid cancer tissue.Methods 90 patients with papillary thyroid car-cinoma(PTC)were selected as the study subjects.During the surgery,PTC tissue and adjacent cancerous tissue were collected at a distance of more than 2cm from the tumor edge.The relative expression level of HULC was detected by real-time fluorescence quantitative polymerase chain reaction(RT qPCR).Transfect TPC-1 cells with different sequences and divide them into Si HULC group,Si Con group,and blank group based on the transfection sequence.The HULC expression level,cell proliferation activity,and invasion ability of the 3 groups were detected.Results The relative expression level of HULC in PTC tissue was significantly higher than that in adjacent tissues(P<0.05).The relative expression level of HULC in PTC cancer tissues with tumor diameter≤2 cm,TNM stage Ⅲ~Ⅳ,and lymph node metastasis was significantly higher than those with tumor diameter>2 cm,TNM stage Ⅰ~Ⅱ,and no lymph node metastasis(P<0.05).Compared with the Si HULC group,the A values of the Si Con group and the blank group at 24,48,72,and 96 hours were significantly higher(P<0.05).Compared with the Si HULC group,the Si Con group and the blank group had significantly higher relative expression levels of HULC and a significantly higher number of inva-sive cells(P<0.05).Conclusion The relative expression of HULC in PTC tissues is high,and it is closely related to tumor size,TNM stage and lymph node metastasis.Only by regulating HULC as early as possible can the proliferation activity and inva-sion ability of TOC-1 cells be effectively reduced.
