Correlation Between miR-223-3p and ECT2 in Esophageal Squamous Cell Carcinoma and Its Effect on Prognosis
Objective To investigate the correlation between miR-223-3 p expression and ECT2 expression in esophageal squamous cell carcinoma and its impact on prognosis.Methods Inclusion of 85 cases of esophageal squamous cell carcinoma with paraffin-embedded tissues from esophageal squamous cell carcinoma and corresponding normal esophageal mucosa tissues.Real-time fluorescence quantitative PCR was used to detect the levels of miR-223-3p and ECT2 mRNA.Pearson correlation coeffi-cient analysis was performed to assess the correlation between miR-223-3p and ECT2 mRNA expression levels.Kaplan-Meier sur-vival analysis and Cox proportional hazards model were employed to analyze the impact of miR-223-3p and ECT2 expression on the prognosis of esophageal squamous cell carcinoma.Results T High expression of ECT2 was observed in 46 cases of esophage-al squamous cell carcinoma,while 39 cases showed low expression of ECT2.The mRNA expression level of ECT2 in esophageal squamous cell carcinoma tissues was higher than that in normal esophageal mucosa tissues,whereas the expression level of miR-223-3p in esophageal squamous cell carcinoma tissues was lower than that in normal esophageal mucosa tissues.The expression levels of ECT2 were correlated with tumor size,differentiation degree,TNM staging,and vascular invasion.Similarly,the expres-sion levels of miR-223-3p were correlated with tumor size,differentiation degree,TNM staging,and vascular invasion.Additional-ly,there was a correlation between miR-223-3p and ECT2 mRNA expression in esophageal squamous cell carcinoma(p=-0.5223,P<0.0001),as well as between miR-223-3p and ECT2 protein expression(x2=21.56,P<0.0001).Kaplan-Meier survival analysis revealed that TNM staging,lymph node metastasis,neural invasion,miR-223-3p expression level,and ECT2 ex-pression level had an impact on the overall survival of esophageal squamous cell carcinoma patients.The Cox proportional hazards model further indicated that TNM staging,lymph node metastasis,neural invasion,miR-223-3p expression level,and ECT2 expres-sion level were associated with overall survival.Conclusion There is a correlation between the expression levels of miR-223-3p and ECT2 in esophageal squamous cell carcinoma.TNM stage,lymph node metastasis,nerve invasion,miR-223-3p expression lev-el,and ECT2 expression level can be used as independent prognostic factors for ESCC.
