Effects of Kaempferol on Proliferation and Apoptosis of Human Renal Epithelial Cells Under High Glucose
Objective To investigate the effects of kaempferol on the proliferation and apoptosis of human renal epitheli-al cells(Hkb20)induced by high glucose,and to clarify its related mechanisms.Methods Human renal epithelial cells were cultured in groups,including normal glucose group(control group),high glucose group,losartan group(positive control)and dif-ferent concentrations of kaempferol groups(10μmol/L,30μmol/L,50μmol/L kaempferol and high glucose co-action).The cell proliferation and apoptosis were detected by CCK-8 method and AV/PI method flow cytometry,the expression of TGF-β1 and Smad3 mRNA was detected by qRT-PCR,and the protein content of TGF-β1 and Smad3 was detected by Western blot.Results High glucose could induce the decrease of human renal epithelial cell proliferation,increase the apoptosis rate,and increase the expression levels of TGF-β1 and Smad3 mRNA and protein(P<0.01).After different concentrations of kaempferol treatment,kaempferol could significantly increase cell proliferation and decrease apoptosis rate.At the same time,the expression levels of TGF-β1 and Smad3 mRNA and protein decreased,and showed a dose-dependent manner(P<0.05).Conclusion Kaempferol could reverse the decrease of human renal epithelial cell proliferation and increase of apoptosis induced by high glucose.This sug-gests that kaempferol may inhibit the decrease of human renal epithelial cell proliferation and increase of apoptosis under high glu-cose environment through TGF-β1/Smad3 signaling pathway,which can eventually lead to renal fibrosis.
