TNF-α调节NF-κB/PXR炎症途径对肺癌细胞生物行为的影响及作用机制

目的 分析TNF-α调节NF-κB/PXR炎症途径对肺癌细胞生物行为的影响及作用机制.方法 将A549细胞分为对照组和干预组,对照组正常培养不做任何干预处理,干预组使用人TNF-α以10 ng/ml浓度共培养12 h;检测细胞增殖、细胞周期、细胞凋亡、细胞侵袭活性;并采用PCR及WB法检测细胞中NF-κB、PXR蛋白和mRNA表达水平.结果 在TNF-α干预24 h、48 h和72 h时,干预组细胞检测的吸光度值明显高于对照组,且差异存在统计学意义(P<0.05);采用TNF-α干预24 h后干预组A549细胞的凋亡率明显低于对照组,且差异存在统计学意义(P<0.05);干预24 h后采用TNF-α处理的干预组细胞的S期比例明显高于对照组,而G2+M期和G0+G1期细胞比例明显低于对照组,且差异存在统计学意义(P<0.05);干预24 h后采用TNF-α处理的干预组细胞侵袭数明显高于对照组,且差异存在统计学意义(P<0.05);干预24 h后采用TNF-α处理的干预组NF-κB、PXR蛋白表达水平均明显高于对照组,且差异存在统计学意义(P<0.05);干预24 h后采用TNF-α处理的干预组细胞NF-κB及PXR mRNA表达均明显高于对照组,且差异存在统计学意义(P<0.05).结论 TNF-α可通过调控NF-κB/PXR炎症途径实现对肺癌细胞增殖、凋亡和侵袭的调控.
Effects of TNF-α on the Biological Behavior of Lung Cancer Cells by Regulating NF-κB/PXR Inflammatory Pathway and Its Mechanism
Objective To analyze TNF-α Regulating NF-κ The effect of B/PXR inflammatory pathway on the biological behavior of lung cancer cells and its mechanism.Methods A549 cells were divided into the control group and the intervention group.The control group was cultured without any intervention,and the intervention group was treated with human TNF-α Co cul-ture with 10 ng/ml for 12 hours;Cell proliferation,cell cycle,apoptosis and invasion were detected;PCR and WB methods were used to detect NF in the cells-K B.PXR protein and mRNA expression levels.Results At 24 h,48 h,and 72 h after TNF-α in-tervention,the absorbance values of cells in the intervention group were significantly higher than those in the control group,and the difference was statistically significant(P<0.05);after 24 h of TNF-α intervention,the apoptosis rate of A549 cells in the in-tervention group was significantly lower than that in the control group,and the difference was statistically significant(P<0.05);after 24 h of intervention,the S-phase proportion of cells treated with TNF-α in the intervention group was significantly higher than that in the control group,while the proportion of G2+M and Go+G,phase cells was significantly lower than that in the control group,and the difference was statistically significant(P<0.05);after 24 h of intervention,the number of invasive cells treated with TNF-α in the intervention group was significantly higher than that in the control group,and the difference was statistically significant(P<0.05);after 24 h of intervention,the expression levels of NF-κB and PXR proteins in cells treated with TNF-α in the intervention group were significantly higher than those in the control group,and the difference was statistically significant(P<0.05);after 24 h of intervention,the expression levels of NF-κB and PXR mRNA in cells treated with TNF-α in the intervention group were significantly higher than those in the control group,and the difference was statistically significant(P<0.05).Conclu-sion TNF-α can regulate the proliferation,apoptosis,and invasion of lung cancer cells by regulating the NF-κB/PXR inflamma-tory pathway.
