摘要

目的 筛选子宫内膜癌差异表达的微小RNA(MicroRNA,miRNA),为深入研究miRNA与子宫内膜癌发生和发展的关系奠定基础.方法 应用miRNA表达芯片检测子宫内膜癌组织及正常子宫内膜组织中miRNA的表达谱.采用实时定量PCR法检测两种子宫内膜组织中差异表达的miRNA,初步验证miRNA芯片结果的可靠性.结果 与正常子宫内膜组织比较,子宫内膜癌组织中有14个miRNA表达上调,6个miRNA表达下调.实时定量PCR验证结果与芯片结果一致.结论 miRNA在子宫内膜癌组织与正常子宫内膜组织间存在差异表达,miRNA表达谱分析将为子宫内膜癌的诊断与治疗提供新的依据.

Abstract

Objective To screen the differential expression of miRNA of endometrial cancer(EC)tissues,to provide a basis for further study of the correlation of miRNA with the development EC.Methods miRNA microarray was used to detect the differential expression of miRNA between human EC tissues and normal endometrial tissues.Real-time quantitative RT-PCR was performed to detect the differential expression of candidate miRNA obtained from microarray experiment.Results Compared with normal endometrial tissues,14 miRNA were upregulated,and 6 miRNA downregulated in EC tissues.The results of real-time quantitative RT-PCR were consistent with those of miRNA microarray.Conclusion The aberrant expression of miRNA in this study may be involved in the pathogenesis of EC.
