目的 分析氧化应激相关的LncRNA对肺鳞癌预后的预测价值.方法 从肿瘤基因组图谱(TCGA)数据库中收集转录组数据和相应的LUSC临床信息.筛选肿瘤与癌旁正常组织间OS基因的异常表达,通过一系列生物信息学分析鉴定8个与预后相关的基因,构建预后模型.同时建立了基于风险评分的Nomo图,该图对LUSC具有较好的识别能力.结果 根据8个基因模型中位风险评分,将患者分为高危组和低危组.生存分析表明,这8个与氧化应激相关的LncRNA在LUAD的预后风险模型中相对可靠.功能富集分析结果表明,LUAD与免疫系统异常密切相关.另发现高危组LUAD患者的基质、免疫和评价评分显著较高,提示TME(tumor microenvironment)不同于低危组,有助于临床医生做出更好的决策.结论 构建一个新的氧化应激相关的lncRNAs信号来预测预后.该模型具有良好的预测价值,可指导今后的个体化治疗策略.
Oxidative Stress-related LncRNAs are Potential Biomarkers to Predict the Prognosis and Immune Response of Patients with LUSC
Objective To prove the role of oxidative stress-related LncRNA in lung squamous cell carcinoma.Methods Through TCMSP database,the active components and potential targets of Shiwei Dazao Decoction in the treatment of leukopenia were screened.The PPI network was constructed by combining Pyrrosia Jujube Decoction and leukopenia mined from multiple da-tabases,and the key targets were obtained by interaction,and the GO and KEGG pathways were enriched and analyzed.Molecular docking technology was used to dock the main active components with the core targets.Results According to the median risk scores of 8 gene models,patients were divided into high-risk group and low-risk group.Survival analysis showed that these 8 Ln-cRNA related to oxidative stress were relatively reliable in the prognostic risk model of LU AD.The results of functional enrichment analysis showed that LUAD was closely related to the abnormality of immune system.In addition,we found that the matrix,immu-nity and evaluation scores of LUAD patients in the high-risk group were significantly higher,showed that TME was different from that in the low-risk group,which was helpful for clinicians to make better decisions.Conclusion We construct a new oxidative stress-related lncRNAs signal to predict the prognosis.The model has good predictive value and can guide individualized treatment strategies in the future.
