Predictive Value of Expression of MACC1 and KIF23 in Cancer Tissues for Postoperative Recurrence of Early Colon Cancer
Objective To observe the expression of colon cancer metastasis-related gene-1(MACC1)and kinesin family member 23(KIF23)in cancer tissues of patients with early colon cancer,and investigate their predictive value for postoperative recurrence of early colon cancer.Methods 80 patients with early colon cancer who were scheduled to receive laparoscopic com-bined fibercolonoscopy were selected,and the mRNA expressions of MACC1 and KIF23 in the cancer tissues and adjacent tissues of the patients were determined by real-time fluorescence quantitative RT-PCR.The mRNA relationship between MACC1 and KIF23 was analyzed by Pearson correlation.After 2 years of postoperative follow-up,patients were divided into recurrence group and non-recurrence group according to whether recurrence occurred.The preoperative basic data of the 2 groups and the mRNA expression of MACC1 and KIF23 in cancer tissues were compared,and a Logistic regression model was established to screen the influencing factors of postoperative recurrence of early colon cancer.ROC curve was drawn to analyze the value of MACC1 and KIF23 mRNA expression in predicting early colon cancer and postoperative recurrence.Results The relative expression levels of MACC1 mRNA and KIF23 mRNA in preoperative cancer tissues were higher than those in paracancer tissues(P<0.05).Pearson correlation showed that the relative expression of MACC1 mRNA and KIF23 mRNA in preoperative cancer tissues was positively correlated(γ=0.326,P<0.05).The proportion of tumor diameter ≥3 cm,obstruction,TNM stage Ⅱ and MACC1 mRNA and KIF23 mRNA in recurrent group were higher than those in non-recurrent group(P<0.05).Logistic regression analysis showed that tumor diameter ≥3 cm(OR=3.947,95%CI=1.256~12.409),obstruction(OR=4.385,95%CI=1.273~15.101),TNM stage Ⅱ(OR=4.071,95%CI=1.058~15.666),MACC 1 mRNA high expression(OR=2.485,95%CI=1.126~7.325),KIF23 mRNA high expression(OR=3.467,95%CI=1.298~9.415)was an independent risk factor for postoperative recurrence of early colon cancer(OR>1,P<0.05).ROC curve found that,AUC of MACC1 mRNA,KIF23 mRNA and their combination in predicting the postoperative recurrence of early colon cancer were 0.771(95%CI:0.649~0.894),0.849(95%CI:0.722~0.976),and 0.928(95%CI:0.825~1.000).Conclusion The expression of MACC1 and KIF23 in cancer tissues of patients with early colon cancer is high,and the combination of the 2 can effectively predict early colon cancer and postopera-tive recurrence.The expression of MACC1 and KIF23 in cancer tissues of patients before surgery should be closely monitored clin-ically to screen high-risk postoperative recurrence patients.
