目的 探讨NCAPG基因对乳腺癌细胞MCF-7 和MDA-MB-231 多项生物功能学作用.方法 分别在MCF-7 及MDA-MB-231 细胞中构建具有siRNA的NCAPG低表达的细胞模型,采用实时的定量PCR法(qRT-PCR)、CCK-8 技术、Transwell实验和流式细胞仪,检测NCAPG在乳腺癌细胞中的生物功能学作用.结果 NCAPG低表达可使乳腺癌细胞生长减慢,转移和侵袭功能减弱.结论 下调NCAPG可减慢乳腺癌细胞增殖和转移的速度,抑制乳腺癌进展.
Abstract
Objective To investigate the effects of NCAPG gene on multiple biological functions of breast neoplasm cells MCF-7 and MDA-MB-231.Methods The cell models with NCAPG low expression with siRNA were constructed in MCF-7 and MDA-MB-231 cells,severally.qRT-PCR,CCK-8 assay,the transwell assay and flow cytometry were used to detect the effect of NCAPG on the cell biology of breast neoplasm.Results Low expression of NCAPG can slow down the growth of MCF-7 and MDA-231 cells,and reduce the metastasis and invasion of MCF-7 and MDA-231 cells.Conclusion Down-regulation of NCAPG can inhibit various biological functions of breast neoplasm cells such as cell growth and metastasis,thereby restraining breast neo-plasm progression.
