摘要
目的 基于网络药理学探究糖尿病足(Diabetic foot,DF)应用湿润烧伤膏(Moist exposed burn ointment,MEBO)治疗的作用机制.方法 采用中药系统药理学分析系统(TCMSP)收集整合MEBO的化学成分及有效成分靶点信息,借助UniProt数据库统一处理靶点基因名称;通过人类基因综合数据库(Gene Cards)分析DF的相关靶点基因,利用Venny 2.1软件分析MEBO活性成分靶点与DF靶点的相关性,获取潜在靶点.利用STRING网站分析交集靶点,随后利用Cytoscape 3.10.0软件构建PPI网络图,找出关键靶点基因.结果 MEBO的主要成分包含黄连、黄柏、黄芩、黄麻苷A等;关键作用靶点为表皮生长因子受体(EGFP)、白细胞介素-1B(IL-1B)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF)等;关键作用通路为VEGF、AGE-RAGE、HIF-1等.结论 经研究MEBO的有效化学成分能够从多靶点、多通路治疗DF,具有促进上皮细胞生长、调节细胞生长与胶原纤维分泌、降低血糖与镇痛作用、保护创面与促进愈合以及减少瘢痕形成等优势.
Abstract
Objective To investigate the mechanism of action of diabetic foot(DF)treated with Moist exposed burn ointment(MEBO)based on network pharmacology.Methods The Traditional Chinese Medicine System Pharmacology Analysis System(TCMSP)was used to collect and integrate the chemical composition and active ingredient target information of MEBO,and the target gene names were unified with the help of UniProt database.Analyzing the relevant target genes of DF through the comprehensive database of human genes(Gene Cards).The correlation between MEBO active ingredient targets and DF targets was analyzed using Venny 2.1 to obtain potential targets.Intersecting targets were analyzed using the STRING website,followed by the construction of a PPI network map using Cytoscape 3.10.0 to identify key target genes.Results The main components of MEBO contain Huanglian,Phellodendron Bark,Scutellaria Baicalensis,Flavoside A,etc.The key targets of action were epidermal growth factor receptor(EGFP),interleukin-1B(IL-1B),interleukin-6(IL-6),and tumor necrosis factor(TNF).The key pathways of action were VEGF,AGE-RAGE,and HIF-1.Conclusion The effective chemical components of MEBO have been studied to be able to treat DF from multi-targets and multi-pathways,with the advantages of promoting epithelial cell growth,regulating cell growth and collagen fiber secretion,lowering blood glucose and analgesic effects,protecting wounds and promoting healing,as well as reducing scar formation.
