Objective To explore the feasibility of a single section dyeing method that can save hexamine silver dye solution. Methods We collected 20 tissue samples from the Pathology Department of Hubei Provincial Hospital of Integrated Chinese and Western Medicine from January 2022 to April 2024,which were pathologically diagnosed as positive for fungal infection. Each sample wax block was sliced into two sections. They were divided into the traditional hexamine silver staining group and the modified hexamine silver staining group, and underwent hexamine silver staining simultaneously. We observed two sets of sections under a microscope and compared the effects of two different staining methods on the results. Results Under the microscope, the modified hexamine silver staining group showed positive staining on the slices.Fungi are black or black brown in color, with clear tissue structure and complete morphology.Compared with the traditional hexamine silver staining group, there was no significant difference in staining effect and diagnostic accuracy. Conclusion The improved single section hexamine silver staining method can ensure the staining effect and effectively save reagents. It is suitable for widespread use in primary laboratories.
