Objective To establish a rabbit model of renal artery stenosis by squeezing the renal artery wall inside the tube,and to evaluate the value of contrast-enhanced ultrasound in the assessment of renal histopathological injury.Methods From December 2023 to March 2024,64 healthy New Zealand rabbits were selected and randomly divided into control group,sham operation group,G1 group(renal artery stenosis rate<50%)and G2 group(renal artery stenosis rate>50%),with 16 rabbits in each group.The rabbit renal artery stenosis model was established by squeezing the renal artery wall inside the tube.On the 2nd day and the 7th day after modeling,8 rabbits in each group were randomly selected for contrast ultrasound examination,blood creatinine and urea nitrogen examination,and renal histopathological examination.Histopathological examination of kidney was performed by HE staining,and pathological quantitative score of renal tubular injury was performed by Jablonski method.The correlation between the parameters of renal cortex contrast-enhanced ultrasound and the pathological score of renal tubular injury was analyzed.Analysis of variance was used to compare the overall difference between groups,and LSD-t test was used for pairwise comparison between groups.T test was used to compare the mean values between two independent samples.Results In this study,the rabbit model of renal artery stenosis was established by squeezing the renal artery wall inside the catheter,tube catheter,and the success rate was 96.7%.Renal artery stenosis>50%can lead to the increase of blood urea nitrogen and creatinine,and the trend of increase is obvious with the prolongation of stenosis time.On the 7th day after modeling,the values were higher than those on the 2nd day(P<0.05).On the 7th day after renal artery stenosis modeling,compared with the 2nd day after stenosis,renal tubule injury was significantly increased in G1 group(P<0.05).Renal tubular injury was significantly increased in the G2 group on the 7th day after renal artery stenosis modeling compared with the 2nd day after stenosis(P<0.05).The results of Pearson correlation analysis showed that WIS,PI,AUC were negatively correlated with the pathological score of renal tubular injury(P<0.05).There was a positive correlation between TTP and pathological score of renal tubular injury(P<0.05).Conclusion It is safe,feasible and successful to establish the model of renal artery stenosis in rabbit by the method of squeezing the renal artery wall inside the tube.Contrast-enhanced ultrasound is an effective adjunct method for noninvasive assessment of renal histopathological injury.
