丹栀逍遥散调控多囊卵巢综合征大鼠胰岛素抵抗的作用机制

目的:研究丹栀逍遥散(DanZhi XiaoYao power,DZXYP)对多囊卵巢综合征(polycystic ovary syndrome,PCOS)大鼠模型胰岛素抵抗的作用及其调控机制.方法:应用皮下埋植左旋18-甲基炔诺酮硅胶棒联合皮下注射人绒毛膜促性腺激素的方法建立PCOS模型,光镜观察卵巢形态及颗粒细胞分布,生化分析仪和电化学发光仪检测血脂、空腹血糖(fasting blood glucose,FBG)和空腹胰岛素(fasting insulin,FINS)等,Western blot检测胰岛素受体底物1(insulin receptor substrate 1,IRS-1)307丝氨酸及AKT 473丝氨酸的磷酸化.结果:DZXYP显著改善PCOS大鼠卵泡的囊性扩张性变化及颗粒细胞排列,与PCOS组相比,DZXYP显著降低TG(P < 0.01)、TC(P < 0.05)和LDL(P < 0.01)水平,显著增高HDL水平(P < 0.05),降低FINS和稳态模型胰岛素抵抗(homeostasis model assessment insulin resistance,HOMA-IR)水平(均P < 0.05),抑制了体重增长(P < 0.05)并减低卵巢重量(P < 0.01),降低了IRS-1的307丝氨酸磷酸化水平(P < 0.01),显著增加AKT 473丝氨酸的磷酸化水平(P < 0.01).结论:DZXYP改善PCOS大鼠胰岛素抵抗的作用可能与降低IRS-1的307丝氨酸磷酸化和增加AKT 473丝氨酸的磷酸化有关.
Effects and mechanisms of DanZhi XiaoYao power on insulin resistance in polycystic ovary syndrome rats
Objective To investigate the effects of DanZhi XiaoYao power (DZXYP) on insulin resistance in polycystic ovary syndrome (PCOS) rats and its regulation on activation of insulin receptor substrate 1 (IRS-1) and Akt. Methods Capsulae Levonorgestreli Silasticus and human chorionic gonadotrophin were used to induce PCOS model in rats. Blood lipid, FBG, FINS, BWGD and OW were measured routinely. Western blot was used to measure activation of IRS-1 and Akt. Results DZXYP ameliorated the cystic expansion of follicles and the disorganized granulosa cells. DZXYP reduced the level of TG, TD, LDL and increased the level of HDL. It decreased FINS and HOMA-IR without effect on FBG. It decreased the BWGD and OW either. DZXYP decreased phosphorylation of IRS-1 307 serine and increased the phosphorylation of Akt 473 serine. Conclusion DZXYP ameliorates the insulin resistance in PCOS rats through decreasing phosphorylation of IRS-1 307 serine and increasing the phosphorylation of Akt 473 serine.
