摘要
目的:制备Vx2接种诱导的兔肾上腺肿瘤动物模型并进行超声影像学观察.方法:将60只新西兰健康免随机等分为接种7 d组、14 d组、21 d组及28 d组,采用直视注射法将VX2肿瘤细胞悬液接种到兔左肾上腺,观察肿瘤影像学表现、接种前后耳正中动脉压变化及高血压发病率、肿瘤平均体积、接种成功率及肿瘤组织形态学.结果:肿瘤接种2周后B超可清晰定位观察肿瘤;接种前后耳正中动脉压均在正常范围,高血压发病率为O;肿瘤总体接种成功率为93.3%;肿瘤体积随接种时间延长而增大,各组肿瘤平均体积差异均有统计学意义(P<0.01).病理检查证实接种2～3周的动物模型最适宜用于肿瘤相关研究.结论:成功制备免VX2肾上腺肿瘤动物模型,该模型制备简便、稳定、成功率高且易于观察,可为肾上腺肿瘤治疗学相关研究提供一个可靠的平台.
Abstract
Objective To establish an adrenal tumor model induced by VX2 in rabbit and observe the ultrasonographic image. Methods Sixty New Zealand rabbits were randomly divided into 4 groups,7-day group, 14-day group, 21-day group and 28-day group. VX2 tumor cell suspension was injected into left adrenal gland of rabbit under euthyphoria to establish an adrenal tumor model. The ultrasonographic images were observed. The median artery blood pressure of rabbit ear in each group was measured before and after grafting and the incidence of hypertension was calculated. Mean tumor volume of each group was calculated. The rate of successful model establishment was calculated.Results Tumor was showed and located accurately by ultrasound imaging two weeks after grafting.The blood pressure in each group was normal. The rate of successful model establishment was 93.3％. The tumor size grew with the increase of time after grafting, which was of statistical significance(P＞0.01). The 14-day model was the best models for tumor study. Conclusion Adrenal tumor model is established successfully in rabbits. It has the advantages of simple procedure, stability and high success rate, and contributes the therapeutic research for adrenal tumor.
