Relationships of polymorphisms of transforming growth factor-β1 and LPA genes with coronary heart disease and in-stent restenosis in the elderly patients
Objective To analyze the polymorphisms of transforming growth factor-β1(TGF-β1)and LPA genes in elderly patients with coronary heart disease(CHD),and to explore their relationships with CHD and in-stent restenosis(ISR)after percutaneous coronary intervention(PCI).Methods A total of 107 elderly patients(>60 years)with CHD underwent PCI in Henan Provincial People's Hospital from June 2020 to June 2022(observation group),and another 107 healthy volunteers underwent physical examination in the same period(control group).The peripheral blood was collected to detect the serum levels of TGF-β1 and lipoprotein a[Lp(a)]by ELISA.The polymorphisms of TGFβ1 and LPA genes were detected by PCR with confronting two-pair primers.The patients in observation group were followed up for 1 year after PCI,and were divided into ISR group(n=33)and non-ISR group(n=74).The gender,age,serum levels of TGF-β1 andLp(a),genotypes and alleles frequencies of TGF-β1 gene rs1800469,rs8105161 and rs2241716 loci,and LPA gene rs3798220 and rs10455872 loci were compared between observation group and control group,and between ISR group and non-ISR group.Results(1)The serum levels of TGF-β1 and Lp(a)were higher in observation group[(13.35±4.77)μg/L,(56.03±11.62)mg/L]than those in control group[(5.76±1.35)μg/L,(34.02± 4.22)mg/L](P<0.05),and there were no significant differences in the gender ratio and age between two groups(P>0.05).The frequencies of CC genotype,CT genotype,TT genotype,allele C and allele T of TGF-β1 gene rs1800469 showed significant differences between observation group(38.32%,38.32%,23.36%,57.48%,42.52%)and control group(20.56%,54.21%,25.23%,47.66%,52.34%)(P<0.05),and those of rs8105161 and rs2241716 showed no significant differences between two groups(P>0.05).The frequencies of TT genotype,TC genotype,CC genotype,allele T and allele C of LPA gene rs3798220 showed significant differences between observation group(55.14%,43.93%,0.93%,77.10%,22.90%)and control group(71.03%,28.97%,0,85.51%,14.49%)(P<0.05),and the frequencies of AA genotype,AG genotype,GG genotype,allele A and allele G of LPA gene rs10455872 showed significant differences between observation group(75.70%,19.63%,4.67%,85.51%,14.49%)and control group(87.85%,11.21%,0.93%,93.46%,6.54%)(P<0.05).(2)The serum levels of TGF-β1 and Lp(a)were higher in ISR group[(16.38±4.78)μg/L,(69.04±9.66)mg/L]than those in non-ISR group[(12.00± 4.13)μg/L,(50.23±6.68)mg/L](P<0.05),and there were no significant differences in the gender ratio and age between two groups(P>0.05).The frequencies of CC genotype,CT genotype,TT genotype,allele C and allele T of TGF-β1 gene rs1800469 showed significant differences between ISR group(57.58%,24.24%,18.18%,69.70%,30.30%)and non-ISR group(29.73%,44.59%,25.68%,52.03%,47.97%)(P<0.05),and those of rs8105161 and rs2241716 showed no significant differences between two groups(P>0.05).The frequencies of TT genotype,TC genotype,CC genotype,allele T and allele C of LPA gene rs3798220 showed significant differences between ISR group(39.39%,57.58%,3.03%,68.18%,31.82%)and non-ISR group(62.16%,37.84%,0,81.08%,18.92%)(P<0.05),and the frequencies of AA genotype,AG genotype,GG genotype,allele A and allele G of LPA gene rs10455872 showed significant differences between ISR group(60.61%,30.30%,9.09%,75.76%,24.24%)and non-ISR group(82.43%,14.86%,2.71%,89.86%,10.14%)(P<0.05).Conclusion The genotypes and alleles frequencies of TGF-β1 gene rs1800469 and LPA gene rs3798220 and rs10455872 are involved in ISR in elderly patients with CHD after PCI,which might be the new targeted therapeutic genes.
