Effect of ophiopogonin on liver regeneration in rats after partial hepatectomy via regulating Wnt/β-catenin signaling pathway
Objective To investigate the effect of ophiopogonin on liver regeneration after partial hepatectomy in rats and the role in regulating Wnt/β-catenin signaling pathway.Methods Forty-two SD rats were randomly divided into sham-operation group,model group and ophiopogonin group,with 14 rats in each group.Ophiopogonin group was injected with 20 mg/kg via tail vein 5 d before operation,while sham-operation group and model group were injected with the same amount of normal saline once a day for 5 consecutive days.On the next day after the last administration,the left lobe and middle lobe of liver were resected in model group and ophiopogonin group.The rats in sham-operation group were only opened the abdominal cavity,and exposed the liver,without resecting the liver.On the 1st and 4th days after operation,7 rats in each group were measured the body mass and liver mass,and the liver index was calculated.The serum glutamic-pyruvic transaminase(GPT)and glutamic-oxaloacetic transaminase(GOT)levels were detected.The serum interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)were detected by ELISA.The relative expressions of proliferating cell nuclear antigen(PCNA),Ki-67,Wnt2,and cytoplasmic and nuclear β-catenin proteins were detected by Western blot.Results The liver indexes were lower in ophiopogonin group[(1.28±0.13)%,(2.01±0.14)%]and model group[(1.22±0.11)%,(1.68±0.14)%]than those in sham-operation group[(2.36±0.15)%,(2.37±0.12)%](P<0.05),and were higher in ophiopogonin group than those in model group on the 1st and 4th days after operation(P<0.05);were higher on the 4th day after operation than those on the 1st day after operation in ophiopogonin group and model group(P<0.05),and showed no significant differences in sham-operation group on the 4th day after operation compared with those on the 1st day after operation(P>0.05).The levels of serum GPT,GOT,IL-1β and TNF-α were higher in ophiopogonin group and model group than those in sham-operation group(P<0.05),and lower in ophiopogonin group than those in model group on the 1st and 4th days after operation(P<0.05);were lower on the 4th day after operation than those on the 1st day after operation in ophiopogonin group and model group(P<0.05),and showed no significant differences in sham-operation group on the 4th day after operation compared with those on the 1st day after operation(P>0.05).The relative expressions of PCNA,Ki-67,Wnt2 and nuclear β-catenin protein were higher in ophiopogonin group and model group than those in sham-operation group(P<0.05),and were higher in ophiopogonin group than those in model group on the 1st and 4th days after operation(P<0.05);were higher on the 4th day after operation than those on the 1st day after operation in ophiopogonin group and model group(P<0.05),and showed no significant differences on the 4th day after operation compared with those on the 1st day after operation in sham-operation group(P>0.05).The relative expressions of cytoplasmic β-catenin protein were lower in ophiopogonin group and model group than those in sham-operation group(P<0.05),and were lower in ophiopogonin group than those in model group on the 1st and 4th days after operation(P<0.05);were lower on the 4th day after operation than those on the 1st day after operation in ophiopogonin group and model group(P<0.05),and showed no significant difference on the 4th day after operation compared with that on the 1st day after operation in sham-operation group(P>0.05).Conclusion Ophiopogonin can reduce the inflammatory response in rats after partial hepatectomy,increase the expression of hepatic proliferation related proteins,and promote liver regeneration,possibly by activating Wnt/β-catenin signaling pathway.
