Correlations of lncRNA MALAT1 with disease activity and inflammatory cytokines in rheumatoid arthritis patients
Objective To compare the expressions of lncRNA MALAT1 in peripheral blood mononuclear cells(PBMC)and fibroblast-like synovial cells(FLSC)of patients with rheumatoid arthritis(RA),osteoarthritis(OA)or traumatic arthritis(TA),and to explore the correlations of lncRNA MALAT1 in PBMC and FLSC with disease activity and inflammatory cytokines of RA patients.Methods From March 2021 to March 2022,17 RA patients(RA group),20 OA patients(OA group),and 17 TA patients(TA group)underwent joint replacement surgery or arthroscopy in Eastern Theater Command General Hospital.Peripheral blood was collected before surgery in three groups to isolate PBMC using Ficoll density gradient centrifugation method,and the synovial tissue samples were obtained during surgery to isolate FLSC using enzymatic digestion.The relative expressions of lncRNA MALAT1 in PBMC and FLSC were detected by real-time fluorescence quantitative PCR,and the levels of interleukin(IL)-2,IL-6 and tumor necrosis factor(TNF)-αwere detected by ELISA.The correlations of the expressions of lncRNA MALAT1 in PBMC and FLSC with the disease activity score in 28 joints(DAS28),erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),rheumatoid factor(RF),anti-cyclic citrullinated peptide antibody(ACPA),IL-2,IL-6 and TNF-α in RA group were analyzed using the Spearman's method.Results(1)The relative expressions of lncRNA MALAT1 in PBMC and FLSC showed significant differences among RA group[0.55(0.45,0.65),0.39(0.33,0.48)],OA group[1.12(0.87,1.34),0.68(0.63,0.76)]and TA group[1.21(1.01,1.46),0.81(0.69,0.98)](H=24.220,P<0.001;H=29.050,P<0.001),and were lower in RA group than those in TA and OA groups(P<0.05).(2)In RA patients,the DAS28 score was 3.450(3.025,4.025),ESR was 29.50(19.75,48.50)mm/h,CRP was 3.700(1.700,7.450)mg/L,ACPA was 53.200(3.075,92.930)u/mL,and RF was 32.000(20.000,105.700)u/mL.The expressions of lncRNA MALAT1 in PBMC and FLSC of RA patients were negatively correlated with the DAS28 score(r=-0.638,P=0.009;r=-0.680,P=0.004)and ESR(r=-0.674,P=0.005;r=-0.508,P=0.047),and not correlated with CRP(r=-0.335,P=0.188;r=-0.341,P=0.211),ACPA(r=-0.309,P=0.387;r=-0.341,P=0.501)and RF(r=-0.355,P=0.231;r=-0.298,P=0.320).(3)The expressions of IL-2,IL-6 and TNF-α showed significant differences in PBMC(H=33.280,P<0.001;H=16.740,P<0.001;H=45.700,P<0.001)as well as in FLSC among RA,OA and TA groups(H=35.310,P<0.001;H=22.130,P<0.001;H=46.130,P<0.001),and were higher in RA group than those in OA and TA groups(P<0.05).(4)The expressions of lncRNA MALAT1 in PBMC and FLSC of RA patients were negatively correlated with the expressions of IL-2(r=-0.520,P=0.035;r=-0.590,P<0.001),IL-6(r=-0.505,P=0.041;r=-0.524,P<0.001)and TNF-α(r=-0.563,P=0.020;r=-0.684,P<0.001).Conclusions The lncRNA MALAT1 expression is down-regulated in PBMC and FLSC of RA patients,which is negatively correlated with disease activity and inflammatory cytokines.lncRNA MALAT1 may be involved in the progression of RA.
