Relationship of expressions of p63 and human epidermal growth factor receptor 2 proteins with clinicopathological features of esophageal adenosquamous carcinoma and their impact on patients'survival
Objective To analyze the clinical significance and prognostic value of the expressions of p63 and human epidermal growth factor receptor 2(HER2)proteins in patients with esophageal adenosquamous carcinoma(EASC).Methods The clinical data of 115 EASC patients diagnosed from December 2005 to December 2021 were retrospectively collected from 800 000 cases in the esophageal cancer clinical diagnosis,treatment and follow-up information database and sample database established by State Key Laboratory of Esophageal Cancer Prevention and Treatment and Henan Key Laboratory for Esophageal Cancer Research of the First Affiliated Hospital of Zhengzhou University.Avidin-biotin-peroxidase complex(ABC)technique was used to detect the expressions of p63 and HER2 proteins.x2 test was conducted to analyze the association between the protein expressions of p63 and HER2 and the clinicopathological characteristics of the patients.Kaplan Meier method was used to analyze the association between the expressions of the two proteins and the prognosis of patients.Multivariate Cox regression was used to evaluate the influencing factors of patients'prognosis.Results In this cohort,the male to female ratio of EASC patients was 3.1∶1,and there was no significant difference in clinicopathological characteristics between male and female EASC patients(all P>0.05).The proportion of patients with family history of tumors and the rate of lymph node metastasis in the p63 pos-itive group were both significantly lower than those in the p63 negative group(both P<0.05).The proportion of male patients and the rate of lymph node metastasis in the HER2 positive group were both significantly higher than those in the HER2 negative group(both P<0.05).The p63 and HER2 positive groups had significantly better survival than the corresponding negative groups(both P<0.05).Multivariate Cox regression analysis found that p63 expression,gender,degree of differentiation,and TNM stage were all independent influencing factors of the prognosis of EASC patients(all P<0.05).Conclusions The expressions of p63 and HER2 proteins are closely related to the prognosis and lymph node metastasis of EASC patients.p63 and HER2 are expected to be prognostic biomarkers and potential therapeutic targets for EASC patients.
