Study on the Mechanism of Early Diabetic Nephropathy Rats and the Effect of Yiqihuoxue Prescription
Objective:To observe the effect of Yiqihuoxue prescription on Ghrelin,AngⅡ,Renin,IL-6 and TNF-α in serum,bronchoalveolar lavage fluid and lung tissue of rats with mechanical ventilation induced lung injury(VILI),and their effects on lung tissue.In order to elucidate the possible mechanism of lung protection.Methods:40 SD rats were randomly divided into blank group,model group,traditional chinese medicine group and positive drug group.VILI rat model was prepared by artificial ventilator after ether anesthesia.The positive drug group was given Ghrelin intraperitoneally before modeling,and the chinese medicine group was given Yiqihuoxue prescription by gavage for 2 weeks after modeling.The lung tissue structure and the degree of inflammatory cell infiltration were observed.The levels of Ghrelin,AngⅡ,Renin,IL-6 and TNF-α in serum and alveolar lavage fluid of each group were detected by ELISA.The expression of IL-6 and TNF-α mRNA in lung tissues was detected by RT-PCR.Results:Compared with blank group,AngⅡ,Renin,IL-6 and TNF-α in serum and lung tissue of rats with mechanical ventilation lung injury model group were increased to varying degrees after modeling,while Ghrelin expression was decreased(P<0.05 or P<0.01).After 2 weeks,compared with the model group,the expression levels of AngⅡ,renin,IL-6 and TNF-α decreased slightly in the chinese medicine group,while the expression of Ghrelin was upregulated(P>0.05);HE staining showed that the morphology and structure of lung tissue were significantly improved in both groups.Conclusion:Yiqihuoxue prescription has a protective effect on lung tissue of VILI rats,which may be related to down-regulating AngⅡ,Renin,IL-6 and TNF-α in lung tissue of VILI rats,up-regulating Ghrelin expression level,and alleviating the injury of lung tissue caused by inflammatory response.
