Analysis of Resource Chemical Components and Extraction Technology of Dietary Fiber in Pueraria Root Residue
Objective Analyze the resource based chemical components in pueraria root residue and examine the optimal process for extracting dietary fiber,in order to provide reference for discovering the value of pueraria root residue resources and improving resource utilization efficiency. Methods The content of polysaccharides and crude fibers in pueraria root residue was determined and analyzed by using the phenol sulfuric acid method and the acid-base digestion method, respectively. In addition, based on a single factor experiment with α-amylase,saccharifying enzyme, and papain were used to prepare dietary fiber, and the swelling force of dietary fiber was measured to determine the optimal process parameters. Results The polysaccharide content in pueraria root residue was 10.30%,and the crude fiber content was 15.58%.The optimal process conditions for enzymatic preparation of dietary fiber were as follows the proportion of composite enzymes(α-amylase,saccharifying enzyme)added was 1:3,the total amount added was 4%,the enzymatic hydrolysis time was 75 minutes, the enzymatic hydrolysis temperature was 65℃,and the total amount of protease added was 0.3%.Conclusion The resource based chemical components such as polysaccharides and crude fibers in pueraria root residue are abundant, and the established dietary fiber extraction process is feasible. The research results can provide scientific basis for the transformation efficiency enhancement and high-value utilization mode of pueraria root residue.
