Experimental study on inhibitory effect of cobra venom enzymes of hospital preparation Sheyao-liquor
Objective To investigate the effect of hospital preparation Sheyao-liquor(Chinese snakebite medicine)on four primary enzyme activities in cobra venom.Methods Using different concentrations of Sheyao-liquor and cobra venom co-incubation as the snake medicinal wine group,while the cobra venom without Sheyao-liquor was used as the snake venom control group,agarose plates were used to compare the inhibitory effects of the Sheyao-liquor group and the snake venom control group on phospholipase A2(PLA2)and fibrinolytic enzyme activities;in addition,the protein hydrolase and hyaluronidase activities of the two groups were detected by for-malin and turbidimetric methods,respectively,to demonstrate the inhibitory effects of Sheyao on protein hydrolase and hyaluronidase in cobra venom.Results The Sheyao-liquor exhibits a robust inhibitory influence on PLA2 in cobra venom at 0.0008,0.004,0.02,0.1,0.5 mg/ml.Moreover,Sheyao-liquor significantly inhibits fibrinolytic enzyme activity at 1.3,13,130 mg/ml.Furthermore,Sheyao-liquor exhibits a pronounced inhibitory effect on proteolytic enzymes in cobra venom when utilized at 0.5,2,8,32 mg/ml.Importantly,the inhibitory impact of Sheyao-liquor on hyaluronidase in cobra venom displays a dose-dependent relationship at 0.0625,0.125,0.25,0.5,1 mg/ml.Conclusion The hospital preparation Sheyao-liquor could inhibit the activity of phospholipase A2,fibrinolytic en-zymes,proteolytic enzymes,and hyaluronidase in cobra venom in vitro.
