Expression and clinical prognostic significance of ANKRD1 in gastric cancer
Objective To screen out the key biomarker ANKRD1 in gastric cancer based on bioinformatics method,and explore its expression and clinical prognostic value in gastric cancer.Methods Differential expression genes of gastric cancer in TCGA data-base were screened in UCSC database,and GO and KEGG enrichment analysis of differential genes were analyzed.Significant prognostic value genes were obtained by univariate COX analysis.UCSC database and RT-qPCR were used to compare the expression difference of ANKRD1 in gastric cancer tissues and normal tissues.GEPIA was used to analyze the relationship between ANKRD1 expression and clinical stage and survival.The correlation between the expression of ANKRD1 and the clinical prognosis of gastric cancer was analyzed by COX regression.GSEA enrichment was used to analyze the signal pathways that ANKRD1 might participate in,and TIMER was used to analyze the relationship between ANKRD1 and immune cell infiltration.Results 1318 differential genes were obtained from the TCGA database,and univariate COX analysis showed that ANKRD1 had independent prognostic value in gastric cancer.The expression of ANKRD1 was significantly up-regulated in gastric cancer tissues(P<0.05);Survival analysis found that the group with low expres-sion of ANKRD1 had a better prognosis(P<0.05);Univariate and multivariate COX analysis showed that ANKRD1 was an independ-ent prognostic factor for gastric cancer(P<0.05);Enrichment analysis showed that ANKRD1 was involved in autophagy regulation and other signaling pathways.Immunoinfiltration analysis showed that ANKRD1 was correlated with macrophage M0 and M1(P<0.05).Conclusion ANKRD1 is highly expressed in gastric cancer,which can be used as an independent prognostic marker of gastric cancer and is associated with immune invasion.
