HPLC法测定颍半夏中10种成分的含量及热图聚类分析

摘要
建立HPLC法测定颍半夏中10 种成分的含量,热图聚类分析栽培颍半夏和野生颍半夏10 种成分的差异.色谱条件如下:采用资生堂C18色谱柱(4.6 mm×250 mm,5 μm);以乙腈(A)-0.01%磷酸溶液(B)为流动相,梯度洗脱;流速1.0 mL/min;双波长检测;柱温30℃;进样量15 μL.结果显示,颍半夏中10 种成分的分离效果较好,方法学考察符合相关要求,热图聚类结果显示颍半夏具有一定的环境聚类特性.该方法为颍半夏质量评价、栽培环境选择等提供依据.
Abstract
A dual wavelength method was established of HPLC for the determination of 10 components in Pinelliae Ternata in Yingshang,and the difference of 10 components in cultivated and wild Pinelliae Ternata in Yingshang was analyzed by heat map cluster.The chromatographic conditions are as follows:Shiseido C18 column(4.6 mm×250 mm,5 μm)was used.Gradient elution was performed with acetonitrile(A)-0.01%phosphoric acid(B)as mobile phase.The flow rate was 1.0 mL/min.Dual wavelength detection was adopted;Column temperature:30℃;the injection volume was 15 μL.The result shows that the separation effect of 10 components of Pinelliae Ternata in Yingshang was better.Methodological investigation meets relevant requirements.The results of heat map clustering showed that wild Pinelliae Ternata had certain environmental clustering characteristics,with a higher content of tar-get components in it.The establishment of this method could provide a basis for quality evaluation and cultivation base selection of Pinelliae Ternata in Yingshang.
