摘要

目的 探讨增加GSTθ表达水平对人类肝癌7721细胞生长及侵袭能力的影响.方法 通过脂质体LipofectAMINE将pcDNA3 -GSTθ真核质粒载体转染获得高GSTθ表达水平的7721稳定转染细胞;采用RT-PCR检测GSTθ mRNA的表达水平;采用细胞记数法测定细胞的生长;采用Boyden小室方法观察细胞侵袭能力的改变.结果 GSTθ高水平表达对7721细胞的生长无明显影响,但使7721细胞的侵袭能力下降.结论 本实验证实了在7721细胞中,GSTθ高水平表达可以降低细胞的侵袭能力.

Abstract

Objective To study the effects of glutathione S-transferase theta(GSTG) on proliferation and invasion of hepatoma carcinoma( HCC) cell line 7721. Methods Overexpresston of GSTO in 7721 cells was performed by stable transfection with a mammalian pcDNA3-GST0 expression vector. GST0 mR-NA were detected by RT-PCR. Cell count assays were applied for evaluation of cell growth. Using Boyden chamber was to used observe the changes of invasion ability. Results The Proliferation of cells earring GST8 gene didn't change. Upexpressron of GST0 could decrease invasive ability of 7721 colls. Conclusion The results demonstrated that GST0 might play an important role in cell invasion in human 7721 cells.
