摘要

目的 探讨辛伐他汀对人肾小管上皮细胞(HKC)上内皮细胞蛋白C受体(EPCR)的影响.方法 采用流式细胞仪检测HKC上EPCR的表达,并将肿瘤坏死因子-α(TNF-α)和辛伐他汀作用于HKC,采用RT-PCR技术对上述因子作用后的HKC上EPCR mRNA表达进行检测.结果 EPCR在HKC中有高表达,TNF-α可下调HKC中EPCR mRNA的表达,而辛伐他汀可以明显改善TNF-α对EPCR的抑制.结论 HKC中EPCR的表达可受到TNF-α的影响,辛伐他汀对上述影响因素有一定拮抗作用,对肾小管上皮细胞损伤可能有保护作用.

Abstract

Objective To investigate the effect of Simvastatin on endothelial protein C receptor (EPCR) in Human Renal Tubular Epithelial Cells (HKC). Methods The expresson of EPCR in HKC was detected by flow cytometry treated by tumor necrosis factor alpha (TNF-a) and Simvastatin. The expression of EPCR in HKC was detected by RT-PCR. Results The results showed that the expression of EPCR were significantly increased in HKC, and the level of EPCR in HKC was suppressed by TNF-a. Simvastatin can obviously reverse the inhibition on EPCR of HKC. Conclusion It was suggested that the expression of EPCR in HKC could be suppressed by TNF-a. Simvastatin may have a protective effect on the renal tubular epithelial cell injury by inhibiting the effect of TNF-a on EPCR.
