摘要

目的 研究携带人IL-2基因腺病毒载体(Ad-IL-24)联用顺铂(DDP)对人非小细胞肺癌NCI-H460细胞生长和凋亡的影响.方法 取对数生长期的NCI-H460细胞,MTT比色法检测Ad-IL-24联合DDP对细胞生长的影响,流式细胞仪分析细胞周期及检测细胞凋亡率.结果 Ad-IL-24联合DDP对NCI-H460细胞的生长抑制作用和诱导凋亡效应均明显优于单用Ad-IL-24和单用化疗药物DDP,且G2/M期细胞比例显著增加(P<0.05或<0.01).结论 Ad-IL-24能提高NCI-H460肺癌细胞对化疗药物DDP的敏感性.

Abstract

Objective To investigate the effects of recombinant adenovirus expressing IL-24 (Ad-IL-24) combined with cisplatin(DDP) on growth and apoptosis of human non-small cell lung cancer NCI-H460 cells. Methods NCI-H460 cells in logarithmic phase were prepared. Effects of Ad-IL-24 combined with DDP on cell growth of NCI-H460 cells were detected by MTT assay. Cell cycle and apoptosis were analyzed by flow cytometry. Results Compared with any single group, the inhibition of cell growth and cell apoptosis in combined group are more effective. Furthermore, Ad-IL-24 combined with DDP significantly increased the ratio of cells in G2/M phase(p<0.05 or <0.01). Conclusion Recombinant adenovirus expressing IL-24 could enhance the sensitivity of lung cancer cells to chemotherapeutic drugs DDP.
