白及多糖琥珀酸酯的抗氧化活性研究

Study on the antioxidant activities of succinated glucomannans from Bletilla striata

目的 研究琥珀酸酯改性白及多糖的抗氧化活性.方法 分别采用Fenton法和邻苯三酚自氧化法测定不同取代度(DS)白及多糖琥珀酸酯BT-2( DS=0.38)、BT-3(DS =0.69)、BT-4( DS=0.85)对OH·和O2-·的抗氧化活性.结果 白及多糖琥珀酸酯对OH·的半数清除率浓度(mg/ml)依次为1.48±0.01( BT-4)、2.65±0.01( BT-3)、2.79±0.02 (BT-2);对O2-·的半数清除率浓度(mg/ml)依次为1.38±0.06(BT-4)、6.44±0.02( BT-3)、21.33±0.13 (BT-2).结果显示,不同取代度白及多糖琥珀酸酯对OH·和O2-·的清除能力大小依次为BT-4> BT-3> BT-2(P<0.05)结论 白及多糖琥珀酸酯的抗OH·和O2-·活性随取代度增大而显著增强.
Objective To study the antioxidant activities of succinated glucomannan from Bletilla striata. Methods Antioxidant activities of succinated glucomannan with different degree of dubstitution (DS) including BT-2 (DS =0. 38) , BT-3 (DS =0. 69) and BT4 (DS =0. 85) against OH ·and O2- · were measured by Fenton reaction and autoxibation of pyrogallic acid. Results The IC50 ( mg/ml) of succinated glucomannans against OH · was measured to be 1.48 ±0.01 (BT-4) , 2.65 ±0.01 (BT-3), 2.79 ±0.02 (BT-2); while their IC50(mg/ml) against O2- · was determined to be 1. 38 ±0. 06 (BT-4), 6.44 ±0.02 (BT-3), 21.33 ±0.13 (BT-2), respectively. Conclusion The antioxidant activities of succinated glucomannans against OH · and O2- · were increased apparently with increasing DS.
