摘要

目的 建立高效分子排阻色谱-蒸发光散射检测法,用于测定右旋糖酐40的分子量及其分布.方法 采用TSK-gel G4000PWXL色谱柱(10 μm,7.8 mm× 300 mm),流动相为水,流速0.6 ml/min,柱温30℃,蒸发光散射检测器.结果 右旋糖酐平均重均分子量在4600~133 800u范围内线性关系良好(R2 =0.991),精密度试验相对标准偏差( RSD)为0.252%,重复性试验RSD为0.460%.结论 该方法灵敏度高,重复性好,可作为右旋糖酐40的分子量及其分布测定的分析方法.

Abstract

Objective To set up a method for determination of molecular weight and distribution of dextran 40 by high performance size exclusion chromatography ( HPSEC) with evaporative light scattering detector (ELSD). Method The TSK-Gel G4000 PWXL (. 10 μm, 7. 8 mm × 300 mm ) column was applied with water as mobile phase. The flow rate was 0. 6 ml/min, and column temperature was set at 30℃. Results As for the average molecular weight of dextran, the linear range was between 4 600 ~ 133 800 u(R2 =0. 991). Relative standard deviations (RSD) of precision and repeatability were 0.252% and 0. 460% respectively. Conclusion The method is accurate with high sensitivity which could be applied for the measurement and quality control of molecular weight and distribution of dextran 40.
