Objective To test the constructine and inducible expression of calreticulin (CRT) on the surface of DC. Method Isolate DC from spleens of C57 mice using fluorescence-activated cell sorting method based on CD11c expression, and analyze the expression of cell surface CRT on inactivated DC u-sing commercialized CRT polyclonal antibody and monoclonal antibody; Purified DC were activated with LPS in vivo for 24h then the inducible expression of cell surface CRT and CD86 were analyzed by flow cy-tometry. Results Inactivated DC express relatively low level of cell surface CRT, and the expression of cell surface CRT and CD86 enhanced when DC is activated by LPS stimulation. Conclusion CRT can be expressed on immature and mature DC, the expression of CRT can increase in cell-surface and body fluidby LPS stimilation, The expression of cell surface CRT on DC may be related with its immunological functions, such as antigen presentation and T cell differentiation.
