摘要
目的 探讨肿瘤坏死因子受体相关因子5 (TRAF5)在大鼠创伤性脑损伤(TBI)中的表达变化及细胞定位情况.方法 利用大鼠锐器切割伤模型,通过Western blot方法检测脑损伤后TRAF5表达的时空变化,免疫组织化学方法检测TRAF5在脑组织中的细胞定位.结果 Western blot检测结果显示,大鼠脑损伤后,TRAF5的表达逐渐升高,损伤后5d达到高峰,此后逐渐降低恢复至正常水平;免疫组化检测结果与Western blot检测结果基本一致.免疫荧光双标记结果显示,TRAF5与星型胶质细胞(GFAP)、神经元(NeuN)之间存在共定位.结论 大鼠脑损伤可以影响TRAF5的表达,这种改变可能参与脑损伤后星型胶质细胞的活化和增殖过程.
Abstract
Objective To investigate the expression of tumor necrosis factor receptor-associated factor 5 (TRAF5) in rat brain after being injuried. Methods The brain stab-injury models of SD (Sprague-Dawley) rat were performed. Western blot and immunohistochemistry methods were employed to detect the expression of TRAF5 at different times after injury. Double immunofluorescent staining was used to observe the distribution of TRAF5. Results Western blot analysis showed TRAF5 level significantly improved at 5 days after injury, and then declined during the following days. The protein expression of TRAF5 was further analysed by immunohistochemistry. Double immunofluorescent staining showed that TRAF5 colocalized with the markers of astrocyte (GFAP) and neuron ( NeuN). Conclusions This is the first description of TRAF5 expression in traumatic brains. These alterations may be involved in the activation and proliferation of astrocytes after brain injury.
