The effect of LINC00943 regulation of miR-331-3p/KMT2A axis on the malignant biological behavior and epithelial mesenchymal transition of cervical cancer cells
Objective:To explore the effect of LINC00943 regulation of miR-331-3p/KMT2A axis on the malignant biological behavior and epithelial mesenchymal transition(EMT)of cervical cancer(CC)cells.Methods:The expression of LINC00943 in normal cervical epithelial cells Ect1/E6E7 and human CC cells lines(HeLa,SiHa,C33A,CaSki)was detected,and HeLa cells was screened as the optimal cell line.The binding sites of miR-331-3p with LINC00943 and KMT2A were predicted.HeLa cells were divided into sh-NC group,sh-LINC00943 group,sh-LINC00943+NC inhibitor group,sh-LINC00943+miR-331-3p inhibitor group,miR-NC group,miR-331-3p mimics group,miR-331-3p mimics+pcDNA group,miR-331-3p mimics+KMT2A group.The proliferation,apoptosis,migration,invasion,and the expression level of EMT related protein were detected in each group.Results:The expression of LINC00943 was significantly upregulated in HeLa cells(P<0.05).LINC00943 targeted to down-regulate miR-331-3p,and miR-331-3p targeted to down-regulate KMT2A.Silenced LINC00943 or overexpression of miR-331-3p inhibited HeLa cells proliferation,migration,invasion,EMT while promoted cells apoptosis(P<0.05).Inhibition of expression of miR-331-3p or overexpression of KMT2A reversed the inhibitory effects of silenced LINC00943 or overexpression of miR-331-3p on HeLa cells proliferation,migration,invasion and EMT(P<0.05).In vivo tumor formation experiments confirmed that silenced LINC00943 significantly inhibited tumor growth and EMT.Conclusions:LINC00943 highly expresses in CC cells,silencing LINC00943 could regulate the miR-331-3p/KMT2A axis to inhibit malignant biological behaviors such as CC cells proliferation,migration,invasion and EMT,resulting in promoting apoptosis.
