Treatment of premature ovarian failure with exosomes from adipose-derived stem cells
Objective:To investigate the effects of adipose-derived stem cell-derived exosome(ADSC-Exo)in treating premature ovarian failure(POF).Methods:ADSC-Exo of rat was prepared.Then ovarian granulosa cells(GCs)were separated and divided into the control group(Ctrl),cisplatin group(Cis)and cisplatin+exosome group(Cis+Exo)according to different treatment strategy.After a certain period of cultivation,the proliferation of GC was compared through CCK8 experiment among three groups.SD female rats were randomly divided into the control group(Ctrl),cisplatin group(Cis)and cisplatin+exosome group(Cis+Exo),with 6 rats in each group.A POF rat model was constructed through cisplatin injection in Cis and Cis+Exo groups,then ADSC-Exo was used to treat these rats for two weeks.The treatment effects were evaluated by the number of follicle count,the levels of FSH,estradiol(E2)and anti-Müllerian hormone(AMH)as well as cell apoptosis.Results:In vitro experiments showed that compared with Cis group,the cell proliferation rate of Cis+Exo group increased 1 day after ADSC-Exo treatment(P<0.05)and was significantly higher after 2 to 4 days of treatment(P<0.001).And the cell proliferation rate of Cis+Exo group was still lower than the control group,and the difference was statistically significant(P<0.05).Animal experiments showed that after 2 weeks of ADSC-Exo treatment,the FSH level was significantly lower in Cis+Exo group than that in Cis group(P<0.001),while the E2 and AMH levels(P<0.001)were significantly higher in Cis+Exo group than those in Cis group(P<0.001).Cis+Exo group showed significant increase in the number of primordial follicles(P<0.05),and decrease in the number of atretic follicles(P<0.05).The apoptosis rate of Cis+Exo group was significantly decreased(P<0.001).Conclusions:ADSC-Exo has a certain therapeutic effect on premature ovarian failure caused by chemotherapy.
