小檗碱对宫腔粘连大鼠的作用及阴道菌群的影响

目的 研究小檗碱(BBR)对宫腔粘连(IUA)大鼠的作用及阴道菌群的影响。方法 取20只6周龄SPF级健康雌性SD大鼠随机分为假手术组、模型组、戊酸雌二醇(EV)组和小檗碱(BBR)组,每组5只,模型组、EV组和BBR组采用机械损伤联合感染的方式建立IUA模型,EV组给予雌激素0。63 mg·kg-1·d-1灌胃,BBR组给予100 mg·kg-1·d-1的BBR生理盐水溶液灌胃,假手术组和模型组分别灌胃等体积的生理盐水溶液,1次/d,各组大鼠均连续给药28 d。收集血清、阴道冲洗液和子宫组织样本,观察各组大鼠的一般情况、子宫形态学和病理学变化,检测各组大鼠的子宫系数、子宫腔通畅性评分、美国生育协会(AFS)评分、子宫内膜腺体数量、内膜厚度、纤维化率、血清炎症因子水平以及阴道菌群情况。结果 与假手术组大鼠相比较,模型组大鼠的子宫系数、纤维化率、子宫通畅性、AFS评分、血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)和IL-6的表达水平、阴道金黄色葡萄球菌和大肠杆菌水平以及Simpson指数均显著升高,子宫内膜腺体数量、内膜厚度、阴道乳酸杆菌和双歧杆菌水平以及菌群丰度、均匀度和Shannon-Wiener指数均显著降低(P<0。05);与模型组大鼠相比较,BBR组子宫系数、纤维化率、子宫通畅性评分、AFS评分、血清TNF-α、IL-1β和IL-6表达水平、阴道金黄色葡萄球菌和大肠杆菌水平以及Simpson指数均显著降低(P<0。05),子宫内膜腺体数量、内膜厚度、阴道乳酸杆菌和双歧杆菌水平以及菌群丰度、均匀度和Shannon-Wiener指数均显著升高(P<0。05)。结论 BBR对IUA模型大鼠具有良好的疗效,经BBR处理后IUA状态明显改善,其机制可能与BBR改善阴道菌群的紊乱有关。
Effect of berberine on intrauterine adhesion and vaginal microbiota of rats
Objective:To investigate the effect of berberine(BBR)on intrauterine adhesion(IUA)and vaginal microbiota of rats.Methods:Twenty SPF-grade healthy female SD rats with 6-week-old were randomly divided into sham group(n=5),model group(n=5),estradiol valerate(EV)group(n=5)and BBR group(n=5).IUA models of the model group,EV group and BBR group were established by mechanical damage combined with infection.EV of 0.63 mg·kg-1·d-1was given intragastric administration to EV group and BBR of 100 mg·kg-1·d-1to BBR group once a day for 28 days.The serum,vaginal washing fluid and uterine tissue samples were collected.The general state,uterine morphology and pathology were observed.The uterine index,uterus patency and American Fertility Society(AFS)scores,the number of endometrial glands,endometrial thickness,the fibrosis rate,expression levels of serum inflammatory factors and vaginal microbiota situation were evaluated.Results:Comparing with the sham group,the uterine index,the fibrosis rate,uterus patency and AFS scores,expression levels of serum tumor necrosis factor-α(TNF-α),interleukin(IL)-1β and IL-6,the content of vaginal staphylococcus aureus and colibacillus as well as Simpson index of the model group were all significantly increased(P<0.05),while the number of endometrial glands,endometrial thickness,lactobacillus and bifidobacteria levels as well as bacteria abundance and uniformity,Shannon-Wiener index were all decreased(P<0.05).Comparing with the model group,the uterine index,the fibrosis rate,uterus patency and AFS scores,expression levels of serum TNF-α,IL-1β and IL-6,the content of vaginal staphylococcus aureus and colibacillus as well as Simpson index of BBR group were all significantly decreased(P<0.05),while the number of endometrial glands,endometrial thickness,the content of lactobacillus and bifidobacteria,bacteria abundance and uniformity,Shannon-Wiener index were all increased(P<0.05).Conclusions:BBR has superior curative effect on IUA model rats,and the IUA state is obviously improved,which potential mechanism might be related with the amelioration of vaginal microbiota disturbance by BBR.
