Determination of Zeaxanthin and Lutein in Lycium Barbarum Extract by HPLC
The chromatographic column Innoval ODS-2(4.6 mm×250 mm,5 µm)is selected with 0.05%triethylamine-acetonitrile as the mobile phase at a flow rate of 0.8 mL/min and a column temperature of 28℃.The samples are determined at 450 nm.The linear relationship of zeaxanthin is good within the range of 0.207 to 20.7 µg/mL,with a determination coefficient R2=1.000 0,an average recovery rate of 113.72%(n=6),a RSD of 1.50%,and a detection limit of 0.027 µg/mL.The linear relationship of lutein is good within the range of 0.216 to 21.6 µg/mL,with a determination coefficient R2=1.000 0,an average recovery rate of 105.96%(n=6),a RSD of 3.78%,and a detection limit of 0.056 µg/mL.The method is fast,simple,stable and accurate.It can be used for simultaneous determination of lutein and zeaxanthin in Lycium Barbarum extract.
